Ansalactam A is a newly discovered ansamycin class polyketide from a marine sediment-derived <i>strepomycete</i> containing a spiro-<i>γ</i>-lactam moiety and a distinctive isobutyl side chain observed for the first time in this class of natural products. Herein we report a novel synthesis of this unique spiro-lactam core and further exploration towards the total synthesis of ansalactam A. The key reaction involves a formal [3+2] cycloaddition which proceeds with high diastereoselectivity to form the core moiety in a single step.
