Pseudorotaxanes belong to a class of supramolecular architectures that have gained popularity in the last few decades due to their general ease of synthesis, ease of modification, low toxicity profiles, and most notably, their ability to “thread” and “dethread” a guest molecule. These systems have been used for a variety of applications ranging from biosensing, <i>in vivo</i> imaging, and drug delivery, to name a few. The pseudorotaxanes generated herein focuses primarily on alkyl-based cationic linear guests, or “threads”, using β-cyclodextrin and γ-cyclodextrin as the hosts due to their ideal cavity sizes, and known ability to enhance certain physiochemical properties of small, hydrophobic drugs. A scope of CD-based pseudorotaxanes have been successfully synthesized and explored via <sup>1</sup>H NMR titrations. The goal of this work is to form ternary complexes using alkylphospholipid-based drugs (APLs), a relatively new class of anticancer agents, to further investigate the viability of a novel drug delivery system.
