Amyloid fibrils are misfolded proteins that self-assemble to form insoluble, elongated, β-sheet structures. These fibrils are linked with the progression of ailments like that of Type II Diabetes, and neurodegenerative diseases like Alzheimer’s and Parkinson’s Disease. A range of molecules have shown the ability to impede the formation of these fibrils. Our work focuses on the inhibitory effect of several compounds (including curcumin and mangiferin) on the formation of amyloid fibrils through the use of Atomic Force Microscopy (AFM) fluid imaging. We have been able to image significant inhibition by these compounds of amyloid fibril formation as well as observe beginning stages and the morphology of fibrils. This work points to potential therapeutic targets in the treatment of amyloid disorders, and a useful technique in imaging fibrils in an environment that more closely resembles <i>in vitro</i> conditions than air dried samples.
