We apply a home-made total-internal reflection fluorescence microscopy to analyze the diffusion of a spherical lipid bilayer (liposome) adhered on a glass-supported lipid bilayer (SLB). We measure the lateral diffusion time of a fluorescent-labelled liposome on a SLB by means of fluorescence correlation spectroscopy. We will report the effects of liposome diameter and ionic strength of solvent on liposome diffusion on a SLB.
