The small-scale purification of mAbs using traditional agarose ProA chromatography media require long residence time to drive adsorption to the resin. Cytvia’s Fibro PrismA chromatography media bypasses this slow-step using convective flow for protein/ligand adsorption. This dynamic property allows for increased flowrates and decreased processing times. An evaluation was conducted to compare the fiber matrix to standard bead-based agarose Protein A media, and the Fibro PrismA decreased total purification time of 100 mL cultures by up to 15X. However, the fiber matrix is prone to occlusion by negatively charged species in cell media, in particular with ExpiCHO, that use traditional sterile filtration steps. The precolumn pressure reaches the max limit in &lt;100 mL causing interruption of the purification method and loss of the material. This abstract will discuss a collaboration with 3M and evaluate their Harvest RC filter for load throughput and clarification of charged contaminants in raw harvest ExpiCHO material. The Harvest RC filtrate reduces occlusion on the fiber matrix across load volumes that would normally cause increased back pressure. The tandem use of the Harvest RC filter with the Fibro PrismA is a path forward to enable high throughput purification in the mg scale to support research and discovery campaigns.
