The TMEM165 gene encodes for a multiple pass membrane protein, localized in the Golgi, that has been linked to congenital disorders of glycosylation.  We have identified the TMEM165 protein as a biomarker for breast carcinoma in a previous glycoproteomic study using patient matched invasive ductal carcinoma and adjacent normal tissues. The TMEM165 protein is not detected in non-malignant breast tissue, increases slightly in early-stage ductal carcinoma in situ (DCIS) cases, and is highly expressed in invasive ductal breast cancer.  Our novel research supports the hypothesis that TMEM165 promotes invasive breast cancer through multiple pathways.  We have used CRISPR/Cas9 knockout of TMEM165 coupled with mutation analysis of the conserved ion transport motifs to determine the role of TMEM165 in invasive breast cancer. Our data indicate that TMEM165 calcium ion antiporter activity represents a new pathway to invasive breast cancer suggesting use as a potential prognostic marker and therapeutic target.
