Lab-on-a-CD devices are centrifugal microfluidic platforms composed by channels and chambers to carry out bio-chemical laboratory tests. Their functions are diverse, but the principle for their fabrication remains in essence constant. Hence, this protocol presentation aims to portray the fabrication of these devices. We describe some known advantages such as portability, low-cost, efficiency, and versatility. Furthermore, we describe some notable applications in several fields with the corresponding variations in the assembly methods employed. We discuss various modifications to the basic assembly methodology that have been applied to achieve different goals or to adapt to new conditions or materials. The examination of these variations provides ideas for every stage of the process: design, cutting, assembly, leak proofing, adhesive check, and sterilization (optional). An in-depth step-by-step guide on how to carry out each stage is established. The time that it would take to complete this fabrication protocol is approximately 3 hours. The difficulty is low, but precision is key to achieve a fully functional device. The incubation of Caco-2 cancer cells and their treatment with cisplatin, an anticancer drug, is highlighted as an exemplary application of the discs to show the type of results that can be obtained. Finally, we provide a troubleshooting guide to overcome the common obstacles in the fabrication process. We hope this serves as a reference to promote the broader utilization of Lab-on-a-CD platforms.
