Sepragen Corporation, based in Union City, California, provides break-through products and processes to the biotechnology and nutraceutical industries for their process development and manufacturing needs Our newest offering goes beyond buffer dilution systems utilizing an integrated “live” conductivity adjustment platform Buffer preparation and conductivity adjustment is a major bottleneck for manufacturing scale purification systems. This bottleneck will become more acute with the greater emphasis and trends to increase production scale throughput by process integration and intensification platforms Sepragen a world leader in bioprocess purification systems has developed and integrated into chromatography systems in-line conductivity blending system that delivers buffers with a fixed conductivity value starting with variable feed streams of concentrated stock solutions The buffer conductivity set point is met by delivery of a selected overall buffer flow rate which will maintain the selected conductivity setpoint by utilizing two selected buffer feed streams with automatic proportionally control driven by Sepragen proprietary software. As an overview the software will read the conductivity source in real time(live) and make ratio adjustments to meet & maintain the target buffer conductivity This system will allow 1.Flexibility, the conductivity the stock buffer does not need to be of fixed value 2. Savings , a manufacturing suite can operate with fewer buffers One or two stocks solution can take the place of several buffers 3. Reduced waste single conductivity( salt) solution can be used over several operations and doesn’t need to be discarded This work will present the Conductivity blending system capabilities from bench scale to a manufacturing scale ~ 60 LPM. Exploring buffer blending abilities over several conductivity targets and various starting buffers . The results(Data) will be presented on Stability, stabilization and scaling up of such buffer preparation along with an overview of the existing commercially available system
