We describe the characterization of an ssDNA aptamer and its application to the development of an electrochemical biosensor for the detection of p57 or Major Soluble Antigen (MSA). This protein is the principal antigen found on the surface of <i>Renibacterium salmoninarum</i> responsible for bacterial kidney disease in salmonids. Aptamers were isolated from a random ssDNA library using GO-SELEX (Graphene Oxide-Systematic Evolution of Ligands by Exponential enrichment) protocol, sequenced and characterized for their binding affinity to the p57 protein. A high affinity aptamer was then immobilized onto stencil printed carbon electrodes using EDC-NHS coupling. Cyclic voltammetry was used for the quantitative detection of the protein which is based on the hindrance of the redox reaction of potassium ferrocyanide on the electrode surface due to the aptamer binding of the target protein. Preliminary results indicate that the aptamer-based sensor can detect the p57 protein over a liner range from 2 to 200 nM.
