The interaction of glycans and glycan-binding proteins (GBPs) plays critical roles in diverse biological processes.<b> </b>It is critical to access user-friendly assays to elucidate the glycan-binding profiles of GBPs and to identify their ligands. We have developed a highly effective multiplex array by immobilizing glycan-bovine serum albumin (BSA) on carboxylate-coated fluorescent magnetic beads (MagPlex beads). Structurally defined homogeneous glycans were synthesized by chemoenzymatic methods and were conjugated to BSA via a homobifuncational squarate linker. The glycan density on BSA can be easily tuned by adjusting the ratio of glycan versus BSA during the conjugation reaction. With this bead-based multiplex array system, the binding profiles of numerous plant lectins, human galectins, and other GBPs were readily obtained. Our array showed advantages in identifying weak binders that may not be able to be identified from some other array system. This platform is robust and flexible for rapid analysis of glycan and GBP interactions and can be easily translated to clinical diagnostics settings.
