Compounds containing α-aminoketone such as bupropion, pyrovalerone, amfepramone, effient are studied as the potential pharmacotherapies for antidepressant and cocaine addiction. The α-aminoketones have not only displayed numerous other biological applications but also been used as versatile intermediates in organic synthesis. Several methods exist to synthesize compounds containing this functional group, only few of them are used for small molecule library preparation. A synthetic protocol was necessary to be developed that could use wide range of readily available starting materials to synthesize diverse array of α-aminoketones. A titanium promoted cross electrophile coupling of imines with Weinreb amides have been developed to synthesize α-aminoketones in moderate to good yield in a chilled reaction.
