The application of metal-based drugs has had a great function as therapeutic agents in the past and in the pioneering times of modern pharmacology, especially as anti-infective agents. Progress achieved in parasite genomics and the identification of a few biomolecular targets hold great promise for the discovery of new ‘mechanism-based’ antiparasitic metallo drugs. The problem of drug resistance posed by micro-organism has resulted in strategies involving modification of active molecules in search for new drug. There is a surge in interest involving combination therapy. This is because the presence of two or more active drug in a single compound will help reduce the pill load, cost, and improve its properties. This research reports the synthesis, characterization and biological studies of metal complexes of two non-steroidal anti-inflammatory drugs: mefenamic acid and indomethacin. The synthetic techniques involved modification of mefenamic acid and indomethacin by incorporating metal ions in the presence of nitrogen donor ligands into the molecular structures of these drugs. Twenty-nine metal complexes have been synthesized so far using some metal salts such as Co(II), Cu(II), Ni(II), and Zn(II) salts. The mefenamato complexes were synthesized by deprotonating mefenamic acid with sodium hydroxide, before reacting with nitrogen donor ligand and metal salts in appropriate solvent while stirring at 40 °C for 1 h. The indomethacin complexes were synthesized by refluxing the ligands and metal salt in acetonitrile:water/ethanol/methanol mixture (4:1 v/v) at 80 °C. The complexes were characterized using physico-chemical techniques such as solubility, melting point, elemental analysis;and spectroscopic techniques such as UV-Vis, FTIR, <sup>1</sup>H and <sup>13</sup>C NMR, CV, EPR, fluorescence; SCXRD and PXRD techniques. Their stability was studied using TG-DSC. They were tested against four bacteria and two fungi.Detailed synthesis, characterization and antimicrobial properties will be described.
