Liquid chromatography forms a key component in modern chemical research and analysis. However, most of this work is based with general separation modes based on adsorption, reversed-phase, normal-phase, ion-exchange, or size-exclusion media.  A more specific approach is employed in affinity chromatography, in which a biologically-related binding agent is instead use for the isolation, analysis, or study of chemicals and biochemicals in complex samples. This presentation will discuss recent developments in the creation and use of analytical applications of high-performance affinity chromatography. Areas to be examined will include chromatographic-based immunoassays, rapid affinity extractions, and multi-dimensional analysis systems for targets such as drugs, hormones, proteins, and environmental contaminants. The use of high-performance affinity columns as a tool for characterizing the binding strength, rates, and interactions of biological interactions will also be considered.  This discussion will include an overview of new developments that have occurred in this field, such as the creation of affinity microcolumns, non-covalent binding agent entrapment, and ultrafast affinity extraction. These developments will be illustrated through applications involving the rapid analysis of biomarkers, chemical bioavailability, and changes in drug-protein interactions during diseases.
