Elevated levels of damaging reactive oxygen species (ROS) are observed in a myriad of disease states, including ischemia/reperfusion events such as heart attacks and strokes. Activation of stress-responsive signaling pathways such as ATF6, a member of the unfolded protein response in the endoplasmic reticulum, promotes tissue viability following ischemia/reperfusion injury. However, chronic ATF6 activation poses risks of toxicity and damaging side effects, motivating the development of prodrugs for disease-targeted ATF6 activation. The design and synthesis of a ROS-activatable ATF6 activator prodrug will be described, as well as its evaluation in primary cardiomyocytes for potential to reduce tissue damage from ischemia/reperfusion injury.
