Immune responses consist of cytokine and cell events with sequencing and duration that follows a natural rhythm of immunity. A growing number of evidences have suggested that the timing of an intervention could either significantly amplify or reduce the benefit of immunotherapeutic agents.<br/><br/>To track the stages of tumor progression and anti-tumoral immune response, we monitored the sentinel organ that bridges the host immune system to the tumor microenvironment - the tumor draining lymph node (tdLN). We investigated the size variation of tdLN as an indicator of innate and adaptive immune responses to inform therapeutic schedule design. With subcutaneously inoculated mouse tumor models, we correlated the size change of the tdLN to cytokine levels in the tumor microenvironment, serum, and both tdLN and contralateral lymph nodes to identify optimal time points to initiate cancer cell death, stimulate dendritic cell activation and T-cell priming.
