While nitrite is used not only in the food industry but also in various chemical reactions such as diazotization to furnish azo-dye compounds. When secondary amine is available in the presence of nitrite, it can form nitrosamine that is a known carcinogen, and due to nitrosamines toxicity, it recently caused the recall of a marketed pharmaceutical product. Detecting residual levels of nitrite, however, can be challenging especially when a number of samples need to be screened. Therefore, more straightforward and user-friendly methods such as colorimetric/fluorometric prove had been developed. In this presentation, a series of nitrite probe molecules will be discussed. Probe molecules were designed to undergo intra-molecular Griess reaction to form triazole product which can be readily detected by color changes. The comparative study was performed with nitrogen-based vs carbon-based nucleophilic probe molecules were prepared and studied by comparative kinetics. With the best performing probe, the authors demonstrated potential application where rapid screening of samples for residual nitrite is needed.
