Dinitrosyl iron complexes (DNICs) are spontaneously generated in mammalian tissue from nitric oxide (NO), biothiols (RSH), and nonheme iron, either labile iron or iron–sulfur clusters. They are readily detected by their characteristic electron paramagnetic resonance signature. However, the endogenous formation as well as the chemical biology of DNICs and the less readily detected dinuclear Roussin’s red salt esters (RSEs) remain but partially understood. In this presentation, we discuss ongoing mechanistic studies of DNIC formation under physiologically relevant conditions.
