Amino acids with additional stereocenters and functional groups present added synthetic complexities to produce. I will describe our efforts to develop biocatalytic routes to efficienctly produce β- and γ-hydroxy non-standard amino acids. We describe the structural and mechanistic properties of two classes of PLP-dependent enzymes that catalyze these reactions. Activity is further imporved through directed evolution to enable g-scale, whole cell catalysis to access these useful compounds.
