Affinity chromatography represents a powerful tool in purifying biologic molecules.  The use of protein A resin for the purification of antibodies has long been recognized as the industry gold standard for the initial chromatographic purification step following clarification of the cell culture harvest.  However, because of the high costs associated with development and use of affinity chromatography resins, they are not commonly pursued unless the target protein of interest is part of a platform technology. In this presentation, we will discuss how understanding the key critical attributes of the target protein can lead to rapid development of affinity ligands and chromatographic purification technologies.  We will demonstrate case studies of how this development process can be applied to standard targets such as IgG antibodies as well as less traditional molecules such as Fabs and fusion proteins.  Furthermore, we will show how these affinity resins when properly utilized in conjugation with subsequent downstream purification steps can lead to improvement in overall manufacturing process efficiency.
