I will discuss a scientific career path that is characterized by the pursuit of distinct classes of nucleic acid chemicals as potential drugs. These classes of chemicals ranging from small to large molecules coincide with where one may practice these discovery efforts: small pharma, big pharma startup and biotech ventures. The vast majority of small molecule drugs can be classified as heterocyclic molecules. Nucleoside drugs, a very mature class of drugs, provide front line therapy for many viral infections and cancer. Antisense Oligonucleotides and newer classes of RNAi oligonucleotides have generated nearly 15 drugs over the past three decades. Nucleoside phosphate mimics, and their prodrugs represent yet another exciting class of drugs promising to overcome some of the deficiencies of nucleosides and oligonucleotides. I will present my drug discovery efforts involving above classes of materials.
