In the presentation we discuss how structural studies have been instrumental in the design of more potent anti-HIV and anti-SARS-CoV-2 inhibitors. We will present structural interactions that are important for the potency of these inhibitors against both wild-type as well as drug-resistant variants. Use of structure from x-ray and molecular simulations in the design of inhibitors and their subsequent structure to activity relationships will be discussed.
