The last decade has witnessed considerable advances in the protein degradation field. Molecular glue molecules co-opting cereblon and leading to cellular protein degradation of transcriptional regulators are clinically approved and referred to as IMIDs. A number of next-generation CELMoDs are either undergoing clinical assessment or are being advanced to the clinic.  This presentation will focus on historical learnings from molecular glue protein degradation programs, applications resulting in successful drug discovery efforts, and current scientific aspects central to ongoing molecular glue drug discovery programs.
