Strain promoted azide-alkyne cycloaddition (SPAAC) reaction is one of the popular 1,3-dipolar cycloaddition reactions, that is utilized in biorthogonal chemistry. Due to high reactivity with dipolar molecules without requirement of metal catalyst, cycloalkynes, in particular cyclooctynes have been employed as a biorthogonal reagent in a biorthogonal chemical reaction to study  biological processes in vivo. The appropriate structure of an alkyne is important to achieve optimum reactivity. Increasing the strain and tuning of electronics in the structure are two main startegies that are followed in general to improve the reactivity of cycloalkynes. However, challenge exists in the designing of optimum cycloalkynes. This draws our interest for the development of novel class of cycloalkynes and investigate their reactivity along with an efficient synthesis. We are focusing on the design of cycloalkynes by incorporating heteroatoms in the ring.
