The ability of T cells to recognize and destroy abnormal cells depends on molecular recognitions at the T cell – target cell interface. Many receptors on T cells are triggered by interactions in trans with their ligands displayed by the target cells. A well-known, but much less understood fact is that some receptors are co-expressed on the same T cells with their ligands, raising the possibility of cell-intrinsic cis-interactions. We and others recently demonstrated that cis-interactions between the immune checkpoint receptor PD-1 and its ligand PD-L1, and between the costimulatory ligand CD80 and PD-L1, regulate the strength of immune checkpoint signaling. In this talk, I will discuss our current understanding on the biology of PD-L1-regulating cis-interactions and toolkits for measuring cis-interactions on the same membranes.
