The focal enzymatic reaction presented in the poster is the ATP-dependent activation of carboxylic acids by conjugation to coenzyme A (CoA) catalyzed by carboxylic acid:CoA ligases (E.C. 6.2.1). In plant metabolism, the reactive thioesters formed by the enzyme are required to fuel many pathways to primary and secondary metabolites and are used by a diverse set of enzymes mediating e.g. acyl group transfer, retro-aldol reactions and redox transformations. Despite their high importance in biotechnology, in vitro (cell-free) access to these co-substrates is still limited. Thus, we aimed to identify novel CoA ligases with improved properties (i.e. high activity, broad substrate scope, acceptance of non-native substrates), which can be used as alternative to existing enzymatic tools in such syntheses. The poster summarizes the characterization of a CoA ligase with respect to the substrate specificity.
