Wee1 inhibition has received great attention in the past decade as a promising therapy for cancer treatment. Therefore, a selective and potent Wee1 inhibitor is highly desirable.  Our efforts to design a safer and more tolerable Wee1 inhibitor led to the discovery of ZN-c3, a highly potent and selective Wee1 inhibitor with balanced ADME and pharmacokinetic properties. ZN-c3 has demonstrated excellent <i>in vitro</i> and <i>in vivo</i> anti-tumor activities in different cell lines as a single agent or in combination. ZN-c3 is currently being evaluated in Phase 2 clinical trials.
