Abstract
Introduction:One of the most common type of injuries sustained during childhood is burn. There are several bandages available for conservative treatment of partial-thickness burns. There isn’t extensive medical literature available about animal testing results regarding the comparison of different treatment methods.
Aim: To develop an adequate testing model, and to compare the four treatment methods of partial-thickness burns
Methods: The authors were examined the healing progress of 104 adult male Wistar rats’s partial-thickness burns developed in their interscapular region. The partial-thickness burns were created by 30 second sustained pressure of a special iron rod, heated up to 130 Celsius. After the development and standardization of the model, the success level of the combination of four different bandages were examined as treatment methods. Biopsy was performed on the 5th, 10th and 22nd day after the burn injuries were developed to assess the depth and range of the burn, the level of epithelialization, and the thickness of scarring. On the 5th day, the % of residually burned surface was determined based on the size ratio of the not epithelialized surface and the burn wound. On the 10th day, the level of wound epithelialization was assessed using a point system (0-open wound, 1-partially closed, 2-closed on multiple levels). On the 22nd day, the scar thickness was measured. The biopsy was performed after hematoxylin eosin was applied. The placement and care of animals was in accordance of ethical regulations and the university’s protocol.
Results: In day 5 and day 10 tsatistical analyses were made by SPSS 22.0 software. Shapiro-Wilk test was used to check normal distribution. A p-value of <0.05 was regarded as significant. Data are presented as mean ± SD in parameters with normal distribution, while median and interquartile values are in the non-normal distribution. Comparisons of four groups were made with Mann-Whitney U tests.
In day 5 we examined the % of residually burned surface was determined based on the size ratio of the not epithelialized surface and the burn wound diameter. We examined 35 rats.
No significant difference was found in the % residually burned surface of between the Aquacel Ag Foam, Curiosa and the combination treatment groups, both treatment methods as Curiosa, Aquacel and the combination treatment were significantly better than Dermazin. We had the lowest % of residually burned surface with using combination treatment. We had the second best result with using Curiosa, after that Aquacel Ag Foam alone, and we had the biggest % of residually burned surface with Dermazin treatment.
On the 10th day, the level of wound epithelialization was assessed using a special point system. Zero score means open wound, score 1 measure the partially closed wound, where the epithelialization is not complete and the score 2 means the section where the epithelialization is closed on multiple levels. We examined 31 rats in day 10. The combination treatment is significantly better than all other treatments. For the tenth day, only fully closed score 2 sections were obtained. This is a huge difference compared to all the other treatments. When using Aquacel Ag Foam and Curiosa, the median is 1 in both cases, but the standard deviation of Curiosa is greater. Significant differences were found between Dermazin and Aquacel Ag Foam, Dermazin and the combination treatment, Aquacel Ag Foam and the combination treatment, and Curiosa and the combination treatment. It can be said that in terms of 10-day score values, the combination treatment proved to be significantly better than all other treatment methods studied.
On the 22nd day, the scar thickness was measured in micrometers. We captured the distance between the epidermis lower cell line and the dermis lower cell line. We examined 38 rats.
Comparisons of four groups were made with Independent Samples t Test in the normal distribution.
At 22 days of follow-up, there was a significant difference in scar thickness between the Dermazin and the combination treatment groups, but no significant difference was found when comparing the other methods. However we can observe that the scar thickness is the thinnest when using the combination treatment.
We had the second thinnest scar when we treated by Curiosa and healed with the thickest scar when it treated by Dermazin.
Conclusion: We can conclude that we found the best method for creating a histologically confirmed and reproducible method for creating partial-thickness burns for animal testing. The burns were created by 30 second sustained pressure of a special iron rod, heated up to 130 Celsius in the interscapular region.
Based on the results of animal testing it can be concluded that the most rapid healing occurred with the combined and same-time use of Curiosa gel and Aquacel Ag foam, making it the most effective treatment.

P.016 Experimental study of the effectiveness of different treatment methods in a rat model of superficial partial-thickness burn injury

We will be pleased to welcome you in Turin (Italy) for the 19th European Burns Association´s Congress in 2022. Italy is honored to hold the meeting of the European Burns Association for the second time, after Verona in 1995. Turin is easily reachable from everywhere in Europe, either travelling by car, train or choosing flight connections. During the meeting, held in the renewed industrial center of the main Italian historical automotive factory (i.e. Lingotto-FIAT), we will meet international burn specialists, focusing on standards of care, progresses in burn treatment, strategies to face disaster situations, with a special attention to education of younger colleagues.
