Crystallization is important in the manufacture of active pharmaceutical ingredients (APIs) to achieve purity and predictable performance for the drug product, while the crystallization of proteins remains a bottleneck in structural biology. Continuous flow manufacturing is increasingly used to improve production efficiencies, and flow crystallization can effectively produce sub-50 micron average crystal sizes with a narrow size distribution by conveniently manipulating the residence time. Short residence times impose constraints on the crystal nucleation kinetics, and DeNovX uses a surface science approach to reduce the kinetic and thermodynamic barriers to nucleation. This presentation will highlight the advantages and challenges in developing flow crystallization approaches at the production and microfluidic scales; the different approaches to achieving the supersaturation required in manufacturing-scale flow crystallization of APIs (e.g., paracetamol cocrystals, ketoconazole, ibuprofen, etc.) vs. microfluidic crystallization of proteins (e.g., lysozyme, BPT, etc.) for structural biology; and the impact of surface energy modified engineered nucleation features on the crystallization outcomes and properties of the crystalline materials.
