Over the past several decades, 10,12-pentacosadiynoic acid (PCDA)-based liposomes have been studied due to their unique colorimetric properties and broad application as sensors. Polymerized liposomes are deep blue, but rapidly turn red when exposed different stimuli. To date, many procedures exist for the preparation of these liposomes on a small lab scale, but many of these methods are difficult to adapt to larger labware or small industrial production. Furthermore, these methods often require the use of organic solvents, which may remain present in the final aqueous suspensions. Herein, we demonstrate a method that avoids organic solvent use during PCDA liposome formation and that has been utilized at multiple scale batches using an inline homogenizer. This method is a viable, greener alternative to the standard thin film rehydration or solvent injectin methods typically used today and results in narrow polydispersities. The self-assembly of liposomes prepared by our method and those prepared the thin film rehydration method were monitored by dynamic light scattering and the mass efficiencies of the two methods are compared.
