A wide variety of lanthanides exhibit certain properties that make them valuable for applications in medicinal chemistry. While Gd has applications as an MRI contrast agent, other lanthanides such as Eu, Tb, and Sm have luminescent properties that can be exploited in medicinal imaging. Additionally to diagnostic applications, the radioisotopes 177Lu and 153Sm can be employed as radiotherapeutic agents. To capitalize on these properties, suitable chelators for lanthanides need not only a high binding affinity for lanthanides, but also require functionalities that amplify the desired properties, such as luminescence, and allow conjugation to bio-relevant targeting vectors. We will present our progress on the development and assessment of lanthanide chelators with tuneable properties for optical imaging applications. Employing Cherenkov radiation provides an in situ excitation source. Our focus lies in the investigation of inter- and intramolecular targeted probes and analyte responsive systems.
