The Encoded Library Technology (ELT) platform at GlaxoSmithKline provides the ability to screen large combinatorial libraries on sub-nanomolar scale for affinity to biomolecular targets. This strategy allows for both the identification of small molecule leads for medicinal chemistry progression and assessment of target tractability. The success of the platform relies on the design of high-quality DNA-encoded libraries that are chemically and structurally diverse as well as effective hit qualification and optimization strategies following DNA-encoded library selection. High-throughput chemistry has the ability to impact both of these aspects of the platform. To this end, recent advances in the platform will be discussed and their impact highlighted.
