Our research laboratories have invested in high throughput experimentation (HTE) capabilities as a method of research design to more rapidly and efficiently progress drug discovery and development.  This poster will describe the application of HTE methods using data-rich design cycles through integration of predictive <i>in silico</i> tools, parallel synthesis, rapid expansion of chemical diversity via nano-scale direct-to-biology (DtB) libraries as well as purified microscale libraries and application of CH arylation chemistry to enable efficient late-stage structure-activity relationship (SAR) exploration. Cross-functional collaborative efforts, both internal and external, enabled a more rapid lead progression.  This disclosure will highlight the use of HTE capabilities as a foundational strategy in a hit-to-lead (H2L) program to discover novel glucosylceramide synthase inhibitors (GCSi) for the treatment of Parkinson’s Disease (PD).
