There is an unmet need for vaccines against fungal infections, causing severe infections in immuno-compromised people. Promising candidates are conjugate vaccines based on cell wall polysaccharide, but the heterogeneous nature of these jeopardize the efficacy of a vaccine. We have synthesized a library of complex but homogenous oligosaccharide structures corresponding to GXM surface polysaccharides from the fungi <i>Cryptococcus neoformans </i>and have used them in screening of mouse monoclonal antibodies and human sera. The results of these screenings as well as immunization of mice using synthetic glycan conjugate vaccines will be discussed. FRET probes of the synthetic glycans have also been produced and their use in the detection and study of mouse mAbs having innate glycosidase activity will be discussed.
