Mammalian glycans on multiple types of glycomolecules are recognized by viruses and bacteria.   These interactions can lead to infection but also are part of normal protein-glycan interactions important in homeostasis in regard to the microbial environments within us.  In addition, humans respond to pathogens by generating specific antibodies to pathogen-derived glycans, as part of the adaptive immune response.  To explore such interactions we have used a variety of approaches including glycan microarrays.  One kind of glycan microarray has chemically defined glycans linked covalently through various chemistries to glass slides.  The other type is one we developed and termed “shotgun” glycan microarrays, that utilized chromatographically-separated natural glycans occurring in glycoproteins and glycolipids from both animals and microbes.  This presentation will focus on discoveries involving virus recognition of glycans, e.g. influenza virus and SARS-CoV-2, antibodies to bacterial and parasitic glycans, e.g., <i>Schistosoma mansoni</i>, as well as new insights into the roles of mammalian mucin glycans in partly regulating bacterial virulence.  These studies are demonstrating the complex interactions between glycans and their recognition in contributing to infections and regulation of microbial interactions.
