Efficient access to complex polycyclic architectures is of paramount importance in the field of natural product synthesis. A rapid synthetic avenue to (+)-Hinckdentine A, a screw-shaped tribrominated marine alkaloid possessing an indolo[1,2-<i>c</i>]quinazoline core embedded in a caprolactam unit bearing consecutive stereogenic centers at C12 and C17a is disclosed herein starting from readily available indoloquinazoline building block. The marine bryozoan <i>Hincksinoflustra denticulate</i>, from which (+)-Hinckdentine A was isolated back in 1987, does not possess much of the alkaloid; therefore, an effective synthetic supply of this natural product is clearly important as biological studies on (+)-Hinckdentine A have not been conducted to date due to its natural shortage.
