There has been little work regarding phthalate replacements, and there remains questions as to whether these replacements have similar/different modes of action or phenotypic outcomes as the phthalate they are replacing. As both classes of compounds are currently on the market, a person/animal could still be exposed to a vast array of phthalate and phthalate replacements simultaneously, creating a complex mixture that, to date, is poorly understood. The objective of this research proposed in this application will incorporate new approach methodologies into a workflow using in silico chemical compound data, existing in vitro data from the EPA CompTox database to group phthalate, phthalate replacements, and their metabolites into clusters to quantitatively evaluate their potential individual and joint toxicity using the zebrafish developmental assay. This study incorporated in-silico and in vitro toxicity profiles of phthalate ester compounds and their replacements for cluster analysis. Using these profiles, we conducted toxicity studies using the zebrafish embryo toxicity assay toxicity study of individual phthalate and phthalate replacements using a zebrafish embryo toxicity assay to identify/verify the in-silico chemical-malformations that are associated with distinct clusters of phthalate and phthalate replacements. Our results demonstrate that phthalate and phthalate replacements have overlapping clusters, that are associated with specific morphologies.
