With the rapid development of biomedicine and nanotechnology, tremendous attention has been paid to stimuli-responsive delivery systems owing to their tumor microenvironment-responsive drug release superiority in tumor therapy. Nevertheless, most drug delivery systems are frequently hampered by their complicated preparation, unsatisfactory sensitivity, and insufficient therapeutic efficacy. It remains a challenge to develop a novel nanosystem to overcome these deficiencies. Herein, we propose a tumor microenvironment-responsive bubble-generating liposomal nanoagent endowed with unique properties including controlled synthesis, desirable stimuli responsiveness, prolonged circulation time and encouraging tumor accumulation. In target tumor region, the gas generator of CO2 or NO encapsulated inside the proposed liposomes can be effectively triggered, resulting in the rapid drug release and significantly improved antitumor efficacy. These findings provide a new perspective for developing stimuli-responsive gas-generating liposomal systems aiming at outstanding therapeutic outcomes for further biomedical application.
