Breast cancer is prevalent in women worldwide and leading cause of death by cancer among women every year. However, there is no effective treatment for breast cancer due to lack of efficient delivery strategy. Here, a thermal ablation strategy combined with immunotherapy is developed to eliminate tumor completely and prevent the metastasis tumor growth. An injectable thermo-responsible Hydrogel was fabricated to induce in situ Nitric oxide (NO) generation, enhancement of crosslinking under NIR laser irradiation. Upon the 808 nm laser treatment, hydrogel turned to form more crosslinking help to release drug slowly and substantially as well as start to release NO to enhance deep drug penetration in tumor site. Moreover, hyperthermia effect and high concentration of NO directly cause cells death by NIR irradiation. Only phototherapy lacks complete ablation of tumor regression. A urgent synergistic therapy is highly required to eliminate tumor completely and cancer metastasis. Immunotherapy showed a potential and most promising candidate to support phototherapy as supporting tool to suppress tumor growth and cancer metastasis. Here, a ligand of the STimulator of Interferon Genes, (STING) agonist and IDO inhibitor were coupled with photothermal therapy. The combinational of phototherapy and immunotherapy synergistically revealed great collaborative efficiency in vitro and suppress the tumor progression in our study. A significant cells toxicity and reduction of tumor growth were demonstrated under this treatment, which could be a promising and effective strategy for future breast cancer treatment.
