<br/>Vancomycin hydrochloride is a well-documented tricyclic glycosylated peptide antibiotic that has been shown to effective against gram-positive bacteria. The present study explores the aggregation and self-assembly of vancomycin in water, phosphate, and borate buffer solutions. UV-VIS absorption spectrum of vancomycin exhibit, λ max at 282 nm. The steady-state emission spectrum in water (excitation wavelength 282 nm) shows an emission maximum centered at 332 nm. Concentration and temperature dependence of absorption and fluorescence studies will be presented. Capillary electrophoresis (CE) chromatogram obtained in borate buffer exhibit multiple peaks, that change in the intensities upon the change in concentration of vancomycin hydrochloride. The CE results of different concentrations of vancomycin hydrochloride in water, borate, and phosphate buffer show possible aggregates of vancomycin hydrochloride with the counterions. A systematic correlation of vancomycin hydrochloride concentrations in water and buffer solutions will be compared using the spectroscopic and CE studies. In addition, Molecular dynamics simulations to determine possible pathways for vancomycin hydrochloride aggregate with the counterions in water will be discussed.
