Deoxy sugars oligosaccharides are a common component of biologically active secondary metabolites.  Glycodiversification, or changing the composition of these glycans, can modulate these natural products activity, offering promise for the development of new therapeutics. The difficulty in synthesizing pure deoxy sugars has hampered efforts to develop glycodiversification into a viable strategy for drug discovery.  Challenges in deoxy sugar oligosaccharide synthesis not only include controlling selectivity in glycosylation reactions, but also developing robust methods for obtaining appreciable quantities of the monosaccharides needed to assemble these molecules.  This talk will detail our labs efforts to develop reliable methods for glycosylation reactions with deoxy sugar substrates, including lessons learned about how stereochemical configuration of different donors and acceptors play in the stereochemical outcomes of these reactions.  Recent examples of the successful application of these technologies to oligosaccharide synthesis and work towards automating these chemistries will also be discussed.
