Polysaccharide conjugated vaccines have benefited billions of people by inducing an adaptive immune response to prevent virulent bacterial infections. Reductive amination is one of the most widely used conjugation chemistries for manufacturing such vaccines. However, due to the complex nature of polysaccharide conjugated proteins, detailed structural characterization of the conjugates has been elusive. Some of the questions to be answered include: Where does activation occur on the polysaccharide, where does conjugation occur on the carrier protein, what is the form of the bond(s) between the polysaccharide and protein once conjugated? The tools used and the data generated to answer these questions will be presented.
