How the cell organises complex biochemical reactions in space is one of the key issues in cellular and molecular biology. Regulating the localisation of reactant components inside separate subcellular compartments is one of the approaches to accomplish this. liquid-liquid phase separation (LLPS) is an alternative option by which compartments can be formed and provide trafficking control within cells. Recently it has been found out a large number of membrane-less organelles (MLOs) are formed through LLPS like nucleoli, Cajal bodies, nuclear speckles, processing bodies (P-bodies), stress granules (SG) organelles etc. However, it is still unclear how MLOs concentrate molecules, maintains and regulate their structures, manage their compositions, and modify internal biochemical activities without interfering with one another within a cell. Therefore, understanding the basic principles of MLO formation via LLPS and comprehending the molecular principles behind the assembly and disassembly process is essential for myriad cell functions and may provide a key to diagnosing aberrant cellular states and aid in the development of innovative treatment techniques. Here, we have investigated the LLPS behaviour of IDP (Mucin) and the factors involved in driving LLPS <i>in-vitro</i> under different physiological and stress conditions. Our results infer that Mucin phase separation is driven by a combination of biotic and abiotic variables rather than merely by electrostatic and hydrophobic interactions.
