Biocatalytic functionalization of <i>sp<sup>3</sup></i> hybridized carbons often requires substrate-specific enzymes or complex cofactors that may not be readily attainable. A promiscuous and straightforward process for biocatalytic oxidation of activated C-H bonds would have wide applications for synthetic and environmental chemistry. Herein we present a novel chemoenzymatic system capable of oxidizing a broad range of benzylic and allylic substrates using molecular oxygen with commercially available enzymes and reagents under mild conditions. Direct oxidation to the carbonyl predominates with minimal formation of under- or over-oxidized products. We show that the system is compatible with multi-enzyme cascade reactions to achieve functionalization beyond oxidation. Additionally, we demonstrate how this chemistry enables opportunities for remediation of polymeric materials and biomass.
