Antibiotic resistance is a serious and growing problem. One enzyme that contributes to the problem is OXA-66, a β-lactamase found in Acenitobacter baumanii, which hydrollyzes carbepenem antibiotics. To carry out 19F NMR experiments on OXA-66, its seven tryptophans were labeled with fluorine using various isomers of fluoroindole. The number of observed peaks and the changes upon binding inhibitors were used to find an optimal labeling strategy.
