Trityl protection has wide applications, especially in a solid-phase synthesis that includes the automatic synthesis of DNA, RNA, and peptides. Trityl protected solid support and nucleoside has been using during the coupling step in DNA oligonucleotide synthesis. Trityl derivatives of amino acids can be used for peptide synthesis. Glycols up to six ethylene glycol units and diols up to decane units were tritylated under controlled and mild conditions. In this process complete consumption of trityl chloride was monitored by TLC then the reaction mixture was extracted to remove excess glycols or diols and pyridinium salt. The majority of ditritylated bi-product and remaining pyridine were removed by precipitation. The tritylated product has a higher yield. Products were analyzed with TLC and ESI and NMR. The product does not require any expensive flash column chromatographic separation.
