Developing versatile, selectiveselective, and efficient catalysts is vital for pharmaceutical chemistry where biaryl moieties are privileged substructures. Due to the demand for drug molecules containing biaryls, chemists have developed a variety of methods to synthesize these compounds, typically using metal mediated cross-couplings, but there remains a significant challenge in using transition metal catalysts in are commonly less efficient in ambient conditions, with green solvents and with with easy catalyst removalare rarely recyclable. Polymeric single chain nanoparticle catalysts have potential as versatile catalysts that can overcome limitations of small-molecule systems: such as provide versatility in catalyst solubility, and recyclability, while also providing the potential for substrate selectivity and lower overall catalyst loading. Herein I have describedevelope polymeric single chain nanoparticle catalysts synthesized using RAFT polymerization with acrylamide-based monomers, and a novel triphenylphosphine derived monomer that forms a Pd(II)-polymer complex that can yield biaryl compounds at a low micromolar catalyst loading through Suzuki-Miyaura couplings under mild ambient oxygen-tolerant conditions. Systematic characterization of different polymer architectures and compositions has revealed relationships between the polymer structure and catalytic efficiency. These polymeric palladium catalysts offer the opportunity for an easily modular macromolecular catalyst system to enable the synthesis of diverse privileged structures using green conditions.
