A sustained-release nanosystem with high drug-loading capacity was developed using open mesoporous silica nanoparticles. High-capacity loading and a sustained release of target drugs was achieved by radially oriented mesopores and a surface functionality. The delivery of dexamethasone was chosen to test this system for sustained anti-inflammatory effects in rheumatoid arthritis (RA). The maximum loading efficiency was confirmed to be ca. 73 wt%, which is about twice that of other silica nanoparticles featuring hexagonal mesopores such as SBA-15. In addition, this system allows a sustained-release profile for 100 h in vitro. Finally, when the nanomedicine was applied to an in vivo RA model, there were significant anti-inflammatory effects over 8 weeks compared with the control groups. The proposed system for nanomedicine delivery could prolong the in vivo concentrations of therapeutic agents and maximize their effect, which should encourage its application.
