For many years my lab has studied various aspects of insecticide toxicology (resistance, novel toxicants, metabolism, etc.) using a variety of approaches (neurobiology, receptor binding, genomics, proteomics, etc.).  Along with improvements in technology there has been greater resolution, but not always greater understanding. For example, understanding of altered target sites as a mechanism of resistance has blossomed, and high resolution of the mutations found lead to a greater understanding of the phenotype and population genetics of the alleles.  In contrast, studies aimed at understanding metabolism-mediated resistance has been bolstered by numerous transcriptomic studies that commonly identify hundreds of candidate genes, but do not always generate a greater understanding of how the phenotype is achieved.  Examples of our changing understanding of the molecular basis of resistance will be presented, along with a set of unanswered questions in insecticide toxicology that may be the highly profitable to investigate in the future.
