The development of enzyme-based catalysis for large-scale manufacture of APIs demands dramatic increases in the speed of enzyme discovery, engineering, and process optimization. State of the art workflows enable low cost, rapid generation, and screening of thousands of enzyme mutants, to identify enzymes with desired properties and eliminate hurdles to portfolio implementation. Prominent example of key intermediate in the commercial synthesis for several APIs are discussed, illustrating both enzyme engineering and catalyst scale up to &gt;1 kg and challenges in developing an industry-leading proprietary enzyme collection.
