Electroactive polymers (EAPs) change their properties in the presence of an electric field. In combination with biorecognition molecules, EAPs can be used as biosensing devices. This presentation will delineate efforts to develop a robust biosensor platform based on aptamer-functionalized EAPs that can be used for the detection of biomolecules. An electrochemical biosensor was used for the detection of two mucin 1 polypeptides. The surface of indium tin oxide slides was modified to enable the electrochemical of growth of a poly(3,4-ethylenedioxythiophene) (PEDOT) and poly{2,2-(3,4-dihydro-2H-thieno[3,4-b][1,4]dioxepine-3,3-diyl)diacetic acid} (ProDOT(COOH)2) copolymer. Three copolymer ratios were investigated to maximize performance: 1:2, 1:1, and 2:1 EDOT : ProDOT(COOH)2. These copolymers were used to covalently attach the mucin 1 aptamer to the biosensing platform to provide the system with target selectivity. The electroactivity of the EAP before and after aptamer attachment in an aqueous electrolyte solution was studied. The attachment of the aptamer to the EAP was confirmed with fluorescence microscopy and cyclic voltammetry. The aptasensor was then used for the electrochemical detection of two mucin 1 polypeptides (short polypeptide: APDTRPAPG, long polypeptide: PDTRPAPGSTAPPAHGVTSA). The limit of detection and stability were evaluated. The musin aptasensor showed stability for at least six days, depending on the ratio of the copolymer, when stored in 0.1 M phosphate buffered saline. The 1:2 EDOT:ProDOT(COOH)2 copolymer was found to be more stable over time with one of the smallest limit of detections, making it the more favorable ratio. The performance of these sensors demonstrated their potential as highly sensitive and selective biosensors for a broad range of analytes.
